The rationale behind this approach is to attach a non-replicative restriction fragment which codes for some identifiable phenotype to a set of restriction fragments, one of which contains an origin of replication. To express its phenotype, the non-replicative fragment must replicate autonomously from the chromosome as a plasmid, so only those fragments attached to an origin-containing fragment can do so.
